Also do a search in the jobs section, lots of info about what companies want from a UI designer.
Step two: Find a game and mod it. Hl2, UT
We used a mix of 3DS Max, Flash, and Photoshop on our most recent project. There were a couple middle-ware apps that we tested and then wound up using. However, those apps were the programmers responsibility and were used for implementation of Flash into the engine.
As for creation tools, Photoshop is a definite but I'd also suggest Flash. For Flash I'd recommend not only learning the art end but learn how to write actionscript as well.
Hope that helps.

tbh if you want to get into ui design you should create your designs in flash. you can do all the little animations like mouse overs, drop down menus, blinking, flashing, sounds etc easily in there. its just the most efficient way to try out different designs. just put a screenshot of the game in the background and create the hud or ui on top of it.
as it has been said before every company approaches ui differently. usually an artist works together with a programmer to get it implemented. there are no standart tools or procedures.
