I'm not familiar with TB but, coldn't it be the 'channel' slot which is marking the 'R' channel instead of 'RGB'? Plus: I think your 'roughness' map is actually 'glossiness'.

I used to think so too, but the "r" channel of a grayscale map is still the same information. What's the rest of your shader look like? Metalness with a black or zero value may help?
Are you being too subtle? Unless those areas are notably lighter or darker I don't think you'll see much... From the limited screencap, It's kinda hard to see enough to tell if this is the expected result.
I don't think this is the issue, either, but have you checked the settings/cog icon and toggled srgb to see if that makes a difference?
