Im tried to start with Substance Painter, but I have a lot of problems with the opacity of the brush and the color picker.. I look for something more focused on hand painting, but i will try again with Substance.

Have to agree with Pyrzern here. For Hand painted assets I also use 3dCoat (it's really close to Photoshop and has also a nice export/import option for photoshop)

What render target were you aiming for with the diffuse texuture?

What quality are you attempting to replicate/emulate? Like an existing character asset.
