What OS do you use? On OSX I'm using Art Director's toolkit, on Win our guys use Picpick. They both maybe a bit heavy if you only want to sample a color (they have rulers, grids and other design related stuff) but there are plenty of simpler pickers like Pixeur, ColorPix, ColorMania, Color Cop and so on.
Photoshop color picker works if you drag&drop from the canvas to other part of the screen. If you opened the color picker from forground/background color you'll get instant feedback.

Lamont, Maya has a colour picker tool that can sample any colour within Maya. If you want to sample something from the desktop I think you're out of luck. I used to load the image in the render view and sample it from there.
