It starts you with the tutorial missions, and you must do those before you do anything else, you cant decline the tutorial missions because they are the essential guide EVE. You will be lost without them. After that you will have to decide what career would you want to pursue in EVE for other guides I use Evelopedia…
@Bacn: You missed out one thing. There is: -PVE -Trade -PI -Mining -R&D (research and development) -SCAMMING (Yes according to the EVE-Pedia, it is legal and there are tutorials for that as well.)

Depends what you want to get into, but you can usually jump into whatever part of the game you want. Planetary interaction, as previously mentioned, is something you can start doing the instant you get your starting money from the tutorial. Trading is easily the best way to make money in the game and all you need for that…
