Yea both brands support ZBrush on all their tablets. I use an XP-Pen screen tablet, works fine with ZBrush. If I were buying a new tablet from XP-Pen atm I'd probably go for the Deco 01, or for Huion maybe the Inspiroy H950P. If you have any concerns about compatibility, you can always experiment with the different brands… -

You should use Weighted Normal modifier before you export it to Substance. That's your issue (in case you still have it in OpenGl channel) -
Mouth modeling for animation
I noticed almost everybody models the mouth closed, lips sealed. The problem I have with this, is that this way makes it more difficult to open the mouth for animation, such as using soft selection to move the upper lip away from the lower one without having the lower lip verts get dragged up too (and vice versa). Also in your example ( creating a polygon that isn't on or result in an open border ) perhaps just using the "fill hole" operation and splitting any edges to complete wour polygon would be easier for you? -
