In xNormal select the "bake hi poly vertex colors" map, and also make sure that "ignore vertex colors" is unchecked on your hipoly mesh.
Have you tried any of the tips here http://www.zbrushcentral.com/showthread.php?t=110022 or looked at the model in another application to see if the UVs are correct?
Have you tried m4dcow's Xnormal suggestion?
Personally I don't know my way around generating textures within zbrush that well, because more times than not problems like this happen and zbrush hides too much of what is going on under the hood for people to easily troubleshoot.
Thats why I suggested xNormal, but hopefully someone that knows a solution within ZBrush can chime in.
1. Have you completely exited spotlight before trying to generate texture from polypaint? (Don't know why this would make a difference, but zBrush is crazy)
2. Make sure in the mesh you started with, that all the faces are sewn together in the UVs, exporting objs sometimes messes things like this up.
Good this is only the diffuse i will sculpt the high details and also make some high diffuse details with other applicatioon and will look awesome . thanks for support.
